Stupid question that I should know the answer but here goes:

- I have a Canoscan 4000 scanner used with Vuescan. I use PS for post
processing. For max accuracy and resolution, should I be scanning RGB
48 Bit mode or RGBI 64 bit mode? I can see the size of the scan file
increase with the I channel, but does it really have any purpose?

BTW, I scan primarily Fuji Velvia slides.

To take advantage of Vuescan's Dust & Scratches filter you need to scan in
RGBI 64 bit mode - Vuescan uses the 4th IR channel to deal with them.

The main use of 64 bit mode is with raw files, so that VueScan can use the
infrared data in its dust/scratch removal filter when you scan from the
raw files. Normally I don't think there's any reason to save the infrared
data in a final output TIFF file. Perhaps some people have special uses
for it.

.... which doesn't mean that you have to switch VueScan to RGBI mode
manually. If the filter is enabled, VueScan will scan RGBI even if the
user interface setting doesn't specify RGBI.
